FLUO 3-AM - Introduction
Fluo-3 AM is a commonly used fluorescent probe for detecting intracellular calcium ion concentration. Fluo-3 AM is a fluorescent dye that can penetrate cell membrane. After entering the cell, Fluo-3 AM can be sheared by esterase in the cell to form Fluo-3, thus being retained in the cell. Fluo-3 free ligand is almost non-fluorescent, and its fluorescence will not increase with the increase of calcium ion concentration. Fluo-3 can be combined with calcium ions, which can produce strong fluorescence with a maximum excitation wavelength of 506 nm and a maximum emission wavelength of 526 nm. The recommended excitation wavelength for actual detection is about 488 nm and the emission wavelength is 525-530 nm. Changes in intracellular calcium ion concentration can be detected using laser confocal microscopy or flow cytometry. & & This Fluo-3 AM (Calcium Ion Fluorescent Probe) is a storage solution prepared in anhydrous DMSO(anhydrous DMSO) with a concentration of 5mM. When used for intracellular calcium ion detection, the common concentration of Fluo-3 AM is 0.5-5 μM. Fluorescent probe loading is usually carried out with an appropriate solution containing 0.5-5 μM Fluo-3 AM and cells incubated at 20-37 ℃ for 15-60 minutes, followed by appropriate washing. After washing, appropriate incubation for 20-30 minutes can be considered to ensure that Fluo-3 AM is completely converted into Fluo-3 in cells. & & Precautions & & AM in this Fluo-3 will solidify and stick to the bottom of the centrifuge tube, the tube wall or the tube cover at 4 ℃, ice bath and other lower temperatures. It can be incubated in water bath at 20-25 ℃ for a moment until it is completely melted. & & fluorescent dyes all have quenching problems, please try to avoid light to slow down fluorescence quenching. & & For your safety and health, please wear lab clothes and disposable gloves.
